Abstract

The designs of many of the experiments in this paper are focused on determining the state of individual cells with respect to the differentiation program. In particular, we are concerned with the moment in time after exposure to inducer at which each individual cell becomes autonomous of the inducer for expression of the differentiation program. The point in time at which a cell becomes irreversibly programmed to the differentiation pathway we operationally define as commitment in this system. The initial experiments presented here outline the methodology we have developed to measure the proportion of cells that have become committed as a function of time and of exposure to inducer. These experiments emphasize the coordinated characteristics of the differentiation program.
